Jayalalitha was very influential, though I don't think she was ever known as a superstar. As for Trisha, though her lead career was longer, her acting skills were never that good enough to be known as a superstar. Even now, her career is practically non-existant. She doesn't get the variety of roles that Nayanthara does, plus she hasn't been able to make a film a success all by herself (none of her women-orientated films worked plus most of them were crap anyway). Nayanthara, on the other hand, has been able to make many heroine-orientated films a hit by herself. Plus, even in hero orientated films, she managed to grab a strong role in most of them, which is quite surprising considering that most hero orientated films only have flowerpot roles for female leads. If I'm not wrong, Nayanthara is known as the Lady Superstar of the Tamil film industry, just as Anushka Shetty for the Telugu film industry.
