It could be but I don't think that SLB offered this role to SRK (and even if, I don't think that he would mean this role)...and anyways, it is a fictionalised historical person (like Jodha Akhbar and Asoka - and MANY other historical characters in movies - got fictionalised, too).
I assume, it's about two characters of two men who live in the presence (or represent someone still living).
If it is about a real life person, it can't be Kabir Singh. Otherwise I would have assumed he is talking about Kabir Singh. But this is a long shot, he is too old for the role.
I don't think its Khilji coz I didn't hear anyone say that Khilji was glorified in the movie. I haven't seen the film. Was he? The promos with Khilji made me cringe so much that I didn't wanna watch it.
Honestly, I am unable to find any good negative roles where the movie did very well as he says. The above 2 are the only ones that come to mind. And they don't seem to fit.
I'm sure that it's neither Khilji nor Kabir Singh (which wasn't even offered to him).
I think I may have got two possible characters I vaguely remember had been written in connection with SRK.
The first one is the host in the Danny Boyle Slumdog Millionaire version of KBC (Anil played this role).
The other one is about the Indian-Vietnamese (alleged???) serial killer Charles Sobhraj who tried (and still does so) to make money with his criminal 'career'.
I'm not sure if indeed SRK got offered the role of Charles in "Main aur Charles" (2015). Wikipedia has a page about him:

I don't think it can be. The interview is from Jan 2017. Padmavat didn't come out till 2018.
The two characters had to be hit movies before this interview. My assumption is he maybe referring to any of the multitude of gangster films made.
Slumdog Millionaire definitively was a very successful film.; Main aur Charles wasn't.
