It was a culmination of a lot of factors. Even during Aniket fight, Sai laments that she has changed a lot - she is not the cheerful girl she used to be who used to sing, dance, laugh. This was all courtesy - the harassment she faced at CN.
The Ladakh fiasco was a huge dent. It shook her. It gave her a message that she doesn't have any right on Virat - a message given by the elders themselves. Whatever she tolerated was because of Virat - when Virat himself seems not hers, it pushes her deeper into anguish. After that was hand-holding where she got to see the ugly side of Virat (as well as the sadistic nature of her in-laws who enjoyed her pain), then ousting in which her self-respect was crushed, the fight she had to put up just to see her own husband at the hospital, insults hurled at her including opportunist, gold-digger, nurse etc. Then Harinee birthday. The Virakhi pool side conversation was a bolt from reality....it was the crucial point from which SaiRat relationship never recovered. It convinced Sai that Virat could never be hers. As @asmi pointed out, since then, Sai has been acting very polite with Pakhi. Her guilt trip started where she started feeling like the third wheel - the thorn that came between soulmates Virakhi. She put her heart into complete freeze mode.
The fact that Virat can never be hers pushes her over the edge - she loses a part of her existence (as Neerja so beautifully put it).

Sai has been made to feel like the third wheel always but after the pool side conversation, it was no more her assumption or observation, it started to look like the truth. Also, after the confrontation Pakhi has become more bold and had started to accuse Virat and Sai without any hesitation and fear. When a person acts without any fear, it sometimes can look like they are able to do it only because they are right and that’s why they are so bold.
When she is being made to think and believe that Virat can’t be hers, she couldn’t enjoy this trip, couldn’t talk to him or be around him in her usual self. She just doesn’t know how to act around him anymore. She tries to act formal with him whenever possible and when she is about to get carried away, wears a mask of acting rude and tries to push him away.
