Ohhh..!!!! lolzzz...dats really strange that it has not yet been made clear in the show about their relationship...the way rajmaata hates nimrit...and the way in last epi amrit was shouting on nimrit it didn't seem like they were sisers at all...

Answer to original question: so far, all indicators show that they are biological sisters. Nimrit seems to be, like her mother, a gentle and sweet person. Even her name "Nimrit", a version of "namrata", means humility. Amrit is all fire, like her grandmother, and prides herself on her royal blood like her father. The title of the show indicates that Amrit will undergo "manthan" or turmoil (literally churning). The negativity in her will rise to the surface first, and in the end, her better/ purer self will prevail.
