Mishti is Ishaan's daughter, Mauli could have said that Mishti is her and Ishaan's daughter.

I don't think Kunal is just a sperm donor. Mishti was conceived when him and Mauli were in love otherwise all women would call their boyfriends/husbands sperm donors when sperm donors are when you want a child but haven't found a partner e.g ekta kapoor.
