Now, after i read the update today. Did or does Mayank's mom know Mayank confessed his love to Nupur? or either way..?

i think she knows .... after nupur leaves ,she says "isko to kuch pata hi nahi hai aur yeh(pointing to mayank) sab kuch jaankar kuch samajhna nahi chahta " ...so its pretty clear that she understood mayank's feelings towards nupur

Yh she understands that Mayank has fallen in love with Nupur, but she also understands and Mayank is indenial!
