Can you take a theme of a novel and employ it in the OS. Like how book-based fanfictions are?

Yes you could, but it would be preferable to write something no one has read. But if you still wish you give in something you've already written it won't be a problem.

Yes ofcourse. There is a difference between plagiarising and inspiration just keep that in mind.
