Hii I wanted to know why a male is playing amma's role..is she actually a transgender in the show?

Yes she is a transgender on the show

Why a male is playing, I don't know but Rajiv sir is a good actor

I think male playing a transgender is more convincing than a female playing such role "shakti" is quiet different but still at times its hard for me to believe rubina as transgender so

It is very very safe for the little girls. Otherwise these two girls would have been the servants or in other dangerous situation if some other normal people gave them refuge.

I think they have purposefully included this character, as a transgender. To show that when the normal sane world discarded the kids, someone who is generally not accepted in the society took them under his/ her wings because the doctor had treated him/ her when no one else helped.
