And when he was unconscious and came back to life aftre was being shot by Shammu, he holds Madhu;s hand and didn't let go. that time he was not awake. So does that mean he subconsciously love her but his ego is so big he wants to go with revenge.

That was the first time I feel the closeness from RK was shown. in the show and the beginning of the love track too.

He dying and holding her hand must be all act (he clearly remembered Mahdubala's words that she said to wake-him-up). He getting shot must not be. Couple of collateral damages on the way to completing a movie.
