i don't think killer is their father. Thols is older (photo) he would have remembered his father's face or if he saw the iron man before. so iron man maybe is a brother or brother in law to Thols's mom. maybe she is from rich family if they go back and claim the sothu so he wanted to eliminate her. people do things for money.
Menaka is the image of her mother so no DNA necesary and he is afraid that she will come back to claim the sothu.

maybe he killed Thols father too.

Something happened and it gave him great pain to go to the extend of wanting to kill Meenu. He is definitely not Thols/Meenu's dad but could have been cheated off his sothu by Meenu's adopted parents which eventually destroyed his own family life thus he wants to destroy Meenu - but somehow he looks afraid of Thol's presence...if he knows Thols is Meenu's brother why is then not going after him too ???....!!😍
