You can cut the video (in Movie maker) but that's not what I do. Go to file => import, video frames into layers. Then select the video you want to use and there would be an option to trim the vidoe, blow where the vidoe appears. Hop I make sense :P

For good quality gifs, you have to reduce the colourfulness of your GIFs as much as you can. Black and White GIFs always has a good quality, if you do not prefer that, you have to refrain from making colorful gifs. You can try some selective colouring as well. :)

P.S.: It depends alot on the quality of the video you use! :)

500 frames are waaay to many frames. In order to have HD looking gifs you should have no more than 50-80 frames per gif.

Use sharpening actions or just sharpen your gif(check out itsphotoshop or yeahps on tumblr for sharpening actions.)
Make sure to not have too many adjustment layers(they lower the quality)
Use 720p or 1020p videos
