To me it seemed Manik lost her the moment she told him that they cannot go back to what they have, he sort of accepted it ,didn't protest the finality of her voice. This meant - his star went far away from him - Nandini and he became all lifeless without her. The hand since the time it connected with her during the 1st scene belonged to her.
The loss is more significant for Nandu because it is his touch that connects directly to his heart. Nandu did gave away her fireflies - the faith in love- so this incident with Manik will make her feel and face that love again.

And watching Nandu caring and worrying for him, Manik is going to realize that she is one star who will always shine for him. His happiness and his love will be restored.

When they both are finally on the same page and word it , that is when the hand will come into play , along with his fireflies.

@Bold: Exactly!!! Which is why, it was it was the defining moment of the episode for me.. not all the 'subsequent' hand drama.. because TBH, that was merely incidental.. even it hadn't happened at that point, they still would've been at a stalemate.. this time she walking away and he was letting her..

Her self respect wouldn't let her stay and without her there would be no one to show him the mirror.. with his voice of reason he would have regressed not reached a point of realization.. unlike how people claim that 'absence makes the heart grow fonder'.. in this case it would've only embittered him because he would've felt like he was being abandoned again, just like before..

The 'hand' incident is nicely circumventing all this and giving her a legitimate reason to be with him and guide him into the light without compromising her dignity, by bringing her compassion into focus..
